Planarians offer a better view of eye development

ScienceDaily (Aug. 2, 2012) ? Planarian flatworms have come under intense study for their renowned ability to regenerate any missing body part, even as adults. But now they may take on a starring role as a model system for studying eye development and eye diseases in vertebrates, including humans.

This expansion of the planarian job description comes courtesy of Whitehead Institute researchers, who this week are publishing in Cell Reports an exhaustive catalog of genes active in the planarian eye.

"It's exciting to get this complete list of genes in one fell swoop," says Whitehead Member Peter Reddien, who is also a Howard Hughes Medical Institute (HHMI) Early Career Scientist and an associate professor of biology at Massachusetts Institute of Technology. "This provides perhaps the most comprehensive list of genes involved in eye biology in a model system other than Drosophila that can be used for rapidly studying the function of those genes."

The compound eyes of fruit flies were the most thoroughly studied invertebrate eyes, owing to the short fly life cycle and a library of well-described eye-related genes in fruit flies. Planarians present a new genetic system, with an eye based on one optic cup lined with pigment cells.

To create a list of genes showing enriched activity in the planarian eye, Sylvain Lapan, a graduate student in the Reddien lab, analyzed more than 2,000 planarian eyes. Lapan and Reddien found 600 active genes and studied 200 of them in more detail. Several of the identified genes are known to have versions that play a role in the vertebrate eye but have not been found in the fruit fly eye. Among these are genes involved in eye development and others associated with age-related macular degeneration and Usher syndrome, a disorder that causes progressive retinal degradation.

One of the key genes identified in planarian eye development is the transcription factor ovo, which activates the expression of many other genes as the eye forms. Until now ovo had been associated with neural tube and germ cell development in other organisms, but not with the eye. In planarians, ovo is vital for eye for regeneration and eye maintenance in the adult, and is also active in eye development in the embryo. In fact, when ovo is experimentally turned off, planarians with head amputations cannot regenerate their eyes and eyes of otherwise normal adult planarians vanish after a couple months.

"Similar mechanisms are used to make eyes during homeostasis, regeneration, and embryonic development in planarians," says Lapan. "We now know way more about genes that regulate eye formation in these animals than for any invertebrate other than Drosophila. Planarian eyes are very different from fly eyes, and we're already seeing the benefits of studying diverse model species, like the discovery of a critical role for ovo."

Clinton challenges Africa to embrace democracy

DAKAR, Senegal (AP) ? In a veiled swipe at China's investments in Africa, U.S. Secretary of State Hillary Rodham Clinton on Wednesday urged African leaders to embrace democracy and partnerships with responsible foreign powers if they want to improve the living standards of their people and address the root causes of extremism that threaten security on the continent.

In a speech in Senegal's capital, Clinton challenged Africa's elite to fully respect human rights and she warned of the consequences of rampant abuses, corruption and intolerance that breed contempt and contribute to instability.

She said America would stand by African reformers and she indirectly took on China. Beijing has been criticized for ignoring human rights concerns, local laws and environmental regulations as it boosts investment in Africa in search of energy and resources to fuel its exploding economy.

"There are still too many places in the region and across the continent where democracy is threatened, where human rights are abused, and the rule of law is undermined," Clinton said. "Too many Africans still live under autocratic rulers who care more about preserving their grip on power than promoting the welfare of their citizens. Violent extremism, transnational crime and rampant corruption all threaten democracy."

The United States, she said, is committed to "a model of sustainable partnership that adds value, rather than extracts it" from Africa. "The days of having outsiders come and extract the wealth of Africa for themselves leaving nothing or very little behind should be over in the 21st century," she said.

Without mentioning China by name, she maintained that unlike other countries, "America will stand up for democracy and universal human rights even when it might be easier or more profitable to look the other way, to keep the resources flowing."

"Not every partner makes that choice, but we do and we will," she said, calling support for democracy and human rights the "heart of the American model of partnership."

Clinton was in Dakar on the first leg of an 11-day African tour that will take her to at least six other nations, including Uganda, South Sudan, Kenya, Malawi, South Africa and Ghana. In Ghana, she will attend the state funeral for the late president, John Atta Mills, who died last week.

Her speech coincided with the first major storm of the rainy season, a positive sign in a place where drought is responsible for a food crisis.

Clinton praised Senegal, the only mainland West African country never to have experienced a coup, for its democratic history and recent elections in which a longtime incumbent lost and handed over power to the victor, Macky Sall, whom she met before the delivering the speech.

But she noted that such trends were not necessarily the norm in the region, such as in Mali or Guinea-Bissau where militaries ousted an elected president.

"Leaders who hold on to power at all costs, who suppress dissent, who enrich themselves, their families and their supporters at the expense of their own people ? who define democracy as one election, one time ? are on the wrong side of history," Clinton said.

In Mali, long considered a model of West African democracy until the coup, Clinton said the military and members of the ousted government must reach consensus to restore civilian leadership and blunt the threat posed by radical Islamists who are taking advantage of a power vacuum in the north.

"We encourage all parties to set aside their differences and work to restore democracy, preserve the territorial integrity of the country, and reject the appeals of violent extremism," she said. She added that the U.S. would continue to withhold full development assistance, including security aid, until a democratically elected government is in place.

Guinea-Bissau, where no president has ever served a full-five year term, Clinton said there was a real threat from what she called the "very troubling trend" of drug trafficking. She warned that unless democracy was restored there was a risk that it could become "totally dependent" on Latin American drug traffickers.

In South Sudan, she will congratulate leaders on reaching the one-year mark of the creation of their country after it split with Sudan. But she also will stress the need for the nascent state to resolve its differences with Sudan that threatened to reignite what had been Africa's longest-running civil war when it ended with a historic peace treaty in 2005.

In Uganda, where the U.S. has sent a small number of special forces troops to help African militaries combat the Lord's Resistance Army of Joseph Kony, Clinton will return to the security theme.

From Uganda, Clinton will travel to Kenya, where in addition to urging leaders to hold peaceful, free and fair national elections in 2013, she will meet Somali officials and underscore U.S. support for completing a planned political transition later in August.

After a brief stop in Malawi, Clinton then heads to South Africa, where she will continue a strategic dialogue with South African officials, promote U.S. business in the country and pay her respects to former President Nelson Mandela, who recently celebrated his 94th birthday.

She will then attend Mills' funeral in Accra, Ghana.

Recycle the molding in the closets of your old house to finish out your renovation project! Sometimes, finding molding to match the molding from old houses can be almost impossible. Never try to use makeshift solutions or inferior equipment to replace tools you need for your home improvement project. While you may be tempted to save time and money this way, the equipment you attempt to replace is expensive or hard to get hold of for a reason. Using stopgap measures can give you unsatisfactory results or even expose you to danger.

Take a close look at your home and ideas. As with any home improvement project, you need to know where to begin. Looking through your entire home and cataloging changes you want to see if you can get the best idea of where to start out. Work through the list from necessity to wants.

If you need to do a project around your house, but don?t have the tools to do it, consider how often you will use the tools before buying. It may be more cost-effective to rent or borrow what you need instead of spending the money to purchase items that will be seldom used.

Try planting a tree to add some more curb appeal. This is a very simple home improvement project. Make sure to consider how big the tree will grow. If you have the space, try to plant two trees to frame your home or your entryway. Planting a tree does not take long at all either.

Changing the upholstery on your furniture will give your room a fresh new look and cost you far less than buying new furniture. It is quite an easy fix for dining chairs and stools but you will likely have to hire a professional to do the work on big pieces, like sofas.

If a squeaky faucet is driving you crazy, chances are you can use this trick to fix the problem: After taking the handle off, detach the faucet stern and apply a thin coating of commercial lubricant to each pair of threads. After you have reassembled it, the faucet should turn on and off smoothly and squeak-free.

When applying a fresh coat of paint to a textured wall or ceiling, use a paint roller with a thick, fluffy nap. These rollers can be more prone to dripping, so you have to be careful. However, they are essential for making sure every nook and cranny on the surface receives some paint.

Talk to neighbors and look for signs of new construction before moving into a new neighborhood. You don?t want to end up living across from the newest McMansion. Find out the chances of the neighborhood changing and what the trends are before buying. The gorgeous hill view from the kitchen might become a new subdivision before you know it.

When it comes to home improvement, never allow a contractor to begin work without having a signed contract first. This is important to ensure that you receive the work that was signed for and have a legal contract to assist you if things do not go according to plan. Be specific and consult with an attorney, if needed.

Although it can be tempting to continually put off home repairs, or hire a professional to do simple repair work and, in doing so, rack up a costly bill, most people are amazed to discover how easily and readily they are able to do basic home repair work and maintenance themselves.
